Comprehensive Analysis of the Effects of Latanoprost on Cultured Human Trabecular Meshwork Cells

Purpose: The addition of latanoprost to trabecular meshwork cells indicates the possibility of a decrease in aqueous humor outflow capacity. In this study, we comprehensively investigated the effects of latanoprost on trabecular meshwork cells by analyzing the expressions of total RNA.
Materials and methods: Total RNA was extracted 24 h after 100 nM latanoprost was added to cultured human trabecular meshwork cells. More than 90 million RNAs from each group were sequenced and their expression frequencies were analyzed.
Results: In total, 2,464 and 3,165 RNAs were significantly upregulated and downregulated, respectively, after the addition of latanoprost to cultured human trabecular meshwork cells. Matrix metalloproteinase (MMP) -3 (952-fold), an extracellular protease matrix, exhibited the maximum increase in expression. Moreover, MMP-1 (392-fold) and VGF nerve growth factor inducible (VGF) (403-fold), a neuroprotective factor, exhibited an increase in expression.
Conclusions: Increased expression of MMPs after the addition of latanoprost to trabecular meshwork cells is suggested to reduce the aqueous outflow capacity. Increased expression of VGF, a neuroprotective factor, might be involved in the neuroprotective effects of latanoprost.
